The town of Plattekill was established in 1849 from part of the town of Shawangunk. In addition to its rural homesteads, the town has the hamlet of Plattekill, a postal hamlet that is focused on the junction of New York State Route 300 and New York State Route 44/55.

Plattekill is at the southern edge of Ulster County, which is part of the New York metropolitan area. The west town line is the town of Gardiner; the town of Shawangunk is on the north; the town of Wawarsing is on the east; and the town of Lloyd is on the south.
