Jerome H. Wood was a local resident and pharmacist who founded the community in the 1840s. It was initially known as "Flatbrook," but a clerical error at the Post Office resulted in the name "Holbrook."

Holbrook has a total area of 12.3 square miles, according to the United States Census Bureau.

Hercules C. Sash House Museum: Holbrook's oldest house, dating to 1820, is now the Holbrook's Historical Society's main museum.

Lake Ronkonkoma: In the early 1800's, a large number of lakes were created in this area for the construction of the Long Island Rail Road.

A 160-acre park that includes athletic fields, a play-structures playground for kids, bocce and shuffleboard courts, basketball and tennis courts, and picnic pavilions. There are also nature paths, a bird-watching area, and a historic tree preserve in the park.

Porky's Plaza: This strip mall's claim to fame is the appearance of its namesake character in Kevin Smith's 1994 film "Clerks."
