Jamesburg was formed as a borough by an act of the New Jersey Legislature on March 19, 1887, from portions of Monroe Township.
The borough was named for James Buckelew, who founded the town in 1852.

The borough form of government used by Jamesburg is one of the original nine types of municipal governments that were established by the New Jersey Legislature in 1897.
Several ethnic groups have settled in Jamesburg, including people of Italian, German, Polish, and Hispanic (Puerto Rican and Dominican) descent. It has a central business district surrounded by a mixture of small single-story shops, several Lutheran churches, a Presbyterian church, a Methodist church, a Catholic church, and a handful of parks.
